ABSTRACT

Objective:To evaluate the role of mitochondrial calcium homeostasis in hyperoxia-induced injury to alveolar epithelial cells typeⅡ(AECⅡ) in rats.Methods:Rat AECⅡcell lines (RLE-6TN) were randomly divided into 3 groups ( n=18 each) using a random number table method: nomoxia group (group C), hyperoxia group (group H), and hyperoxia plus ruthenium red group (group HR). The cell lines were cultured in a conventional incubator for 4 h in group C. The cell lines were incubated in an incubator with 90% O 2 for 4 h in group H. In group HR, ruthenium red 2 μmol/L was added first, and then the cell lines were incubated in an incubator with 90% O 2 for 4 h. After the treatment, the Ca 2+ concentrations in mitochondria were measured using the mitochondrial Ca 2+ specific fluorescent probe Rhod-2-AM, the ROS level was measured using the fluorescent probe DCFH-DA, and the cell morphology and mitochondrial ultrastructure were observed. Results:Compared with group C, the Ca 2+ concentrations in mitochondria and ROS level were significantly increased in H and HR groups ( P<0.05). Compared with group H, the Ca 2+ concentrations in mitochondria and ROS level were significantly decreased in group HR ( P<0.05). Compared group C, the cell volume was reduced, the morphology was shrunken round, the cell arrangement was loose, the mitochondrial double-layer membrane structure was broken, and the mitochondrial cristal fragments were observed in group H. Compared with group H, the cell morphology was significantly improved, and the slight damage to mitochondrial double-layer membrane structure was found, and the structure of mitochondrial cristae was normal in group HR. Conclusion:Mitochondrial calcium homeostasis is involved in the pathophysiological process of hyperoxia-induced injury to AECⅡ in rats.

Objective@#To evaluate the effect of deferoxamine on hyperoxic lung injury in rats.@*Methods@#Twenty-four adult male Sprague-Dawley rats, weighing 170-230 g, were divided into 3 groups (n=8 each) using a random number table method: hyperoxia group (group H), deferoxamine group (group D) and air control group (group C). Mechanical ventilation was performed after tracheal intubation, group C inhaled air, and H and D groups inhaled 90% oxygen.Deferoxamine 50 mg·kg-1·h-1 was continuously infused via the tail vein for 4 h via the tail vein at the same time of mechanical ventilation in group D. The equal volume of normal saline was infused in H and C groups.At 4 h of mechanical ventilation, lungs were removed for examination of pathological changes of lung tissues and for determination of wet/dry weight ratio (W/D ratio) and levels of pulmonary surfactant protein C (SP-C), xanthine oxidase (XOD) and glutathione reductase (GR) in lung tissues and broncho-alveolar lavage fluid (BALF) were detected by enzyme-linked immunosorbent assay.@*Results@#Compared with group C, the pathological score of lung tissues and W/D ratio were significantly increased, the levels of SP-C and GR in lung tissues and BALF were decreased, and the level of XOD in lung tissues and BALF was increased in H and D groups (P<0.05). Compared with group H, the pathological score of lung tissues and W/D ratio were significantly decreased, the levels of SP-C and GR in lung tissues and BALF were increased, and the level of XOD in lung tissues and BALF was decreased in group D (P<0.05).@*Conclusion@#Deferoxamine can inhibit oxidative stress response and alleviate hyperoxic lung injury in rats.

ABSTRACT

Objective To explore the combined detection of B‐type natriuretic peptide (BNP) and glycosylated antigen 125 (CA125) in differential diagnosis of dyspnea .Methods BNP and CA125 were detected and compared among 106 cases with cardiac dyspnea ,56 cases with pulmonary dyspnea and 60 healthy subjects .Results BNP and CA125 levels of dyspnea patients were higher than healthy subjects ,and those of cardiac dyspnea patients were higher than pulmonary dyspnea patients (P<0 .05) .With the ag‐gravation of heart failure ,BNP and CA125 levels gradually increased .Combined detection of BNP and CA125 were with higher diag‐nostic sensitivity and specificity (P<0 .05) .After treatment ,BNP and CA125 levels significantly decreased (P<0 .05) .Conclusion Combined detection of BNP and CA125 could be with high specificity and sensitivity ,helpful for identifying cardiac or pulmonary dyspnea and dynamic observation of the disease .

ABSTRACT

In order to evaluate the moUuseicidal effect of 50% wettable power of niclosamide ethanolamine salt(WPN)combined with urea against Oncomelania snails in the field,4 g/m~2 WPN,4 g/m~2 WPN+20 g/m2 urea and 4 g/m~2 WPN+30 g/m~2 urea were used for mollusciciding with the spraying method.The results showed after 7 days,the mortality rates of snail were 74.43% for 4 g/m~2 WPN,90.32% for 4 g/m~2 WPN+20 g/m~2 urea and 94.83% for 4 g/m~2 WPN+30 g/m~2 urea,respectively.It is indicated that WPN combined with urea can improve the molluscieidal effect significantly.

ABSTRACT

Objective To evaluate the clinical value of CT perfusion (CTP) imaging for providing quantitative information about angiogenesis in patients with lung carcinoma and investigate the correlation of CTP enhancement parameters and histological microvessel density (MVD) with lymphatic involvement in peripheral lung carcinoma. MethodsFifty-three patients with pathology-proved peripheral lung carcinoma underwent CT perfusion scan before operation. The enhancement parameters of CTP were calculated based on the time-density curves (TDC) of fist pass phase. All cases were classified into two groups according to pathologic results: tumor with and without lymph node involvement. Two-sample t test was used for the statistics. The ROC curve was used to assess the efficiency of the enhancement parameters of CT perfusion and MVD for predicting lymphatic involvement.Results Tumors with lymph node involvement had significantly higher value of MVD than those without lymph node involvement (64.69±16.34 and 42.67± 16.78, respectively,t=4.84,P<0.01). Tumors with lymph node involvement had significantly higher value of CTP enhancement parameters (PH, M/A, PV) than those without lymph node involvement [PH= (41.79±15.50) and (29.99±10.91) HU,M/A =0.24±0.09 and 0.15±0.06, PV=(2.14±1.09) and (1.27±0.53) ml·min~(-1)·ml~(-1), t=3.21,3.95, 3.66, P<0.01, respectively]. The CTP enhancement parameters (PH, M/A, PV) of lung cancer correlated positively with the MVD, the highest correlation coefficient was between the PV and MVD (r=0.716, P<0.01). MVD and PV had higher values for predicting lymph node involvement in ROC curve analysis.The sensitivity, specificity and accuracy for predicting lymph node involvement were 80.8%, 81.5% and 81.1% or 84.6% ,85.2% and 84.9% respectively if MVD>52/0.74 mm~2 or PV>1.52 ml·min~(-1)·ml~(-1). ConclusionThe CT perfusion PV and histological MVD have good correlation with lymph node involvement in peripheral lung carcinoma and are important predicting parameters before operation.

ABSTRACT

Objective To investigate the relationship between CT signs of portal hypertension and histopathologic stage of chronic hepatic fibrosis and cirrhosis. Methods Tri-stage enhance volume CT scan of upper abdomen was performed in 84 participants, including 48 patients with hepatic fibrosis confirmed by liver pathologic biopsy which divided into S1 (12/48), S2 (14/48), S3 (9/48) and S4 (13/48),16 patients with typical cirrhosis, and 20 healthy subjects as a control group. Measured the caliber of left and right branch of portal vein, MPV, SV and SMV at MIP images respectively, observed the collateral circulation, ascites and the size of spleen and then studied comparatively these measured parameters of different histopathologic stage. One-Way ANOVA was performed in the comparison of the vascular diameter of portal system and the size of spleen(SNK was used in the comparison between the groups). x2 test ofR × Ctable was performed in the comparison of ascites and collateral circulation among groups, and the vessel of portal system which has the greatest impact on the pathological staging of hepatic fibrosis was investigated with Logistic regression analysis. Results The caliber of left branch of portal vein, right branch of portal vein. MPV. SV and SMV were (0.98±0.11). (1.00±0.12), (1.33±0.11). (0.75±0.10).(1.07±0. 12) em respctively, the size of spleen was (128. 55±30. 56) cm<'3>, and collateral circulation and ascites were not found in control group. SV enlarged gradually in test groups and showed S1 (0. 86±0. 12) cm, S2(0. 96±0. 11) cm, S3(1.07±0.08) cm, S4(1.09±0. 10) cm, typical cirrhosis (1.18±0. 19) cm respotively. The difference between each group of S1 to typical cirrhosis and control group was significant, and the same result was seen among S3 to S4, cirrhosis and S1 to S2. Logostic regression analysis showed that the standardized regression coefficient of SV was maximum (2. 719) and had statistical significance(P <0. 01). The incidence of collateral circulati on and ascites in patients with typical cirrhosiswas significant higher than that of normal liver and every stage of hepatic fibrosis (P < 0. 05). Conclusion CT scan may be helpful for the early detection of advanced hepatic fibrosis or early stage of liver cirrhosis for patients with chronic liver disease.

ABSTRACT

Objective To explore the changes on the value of CVP with different body postures after upper abdominal surgery in order to provide accurate basis for better monitoring of central venous pressure(CVP). Methods The CVP, blood pressure,pulse,respiration of 43 patients after upper abdom-inal surgery with indwelling central venous catheter were measured under supine position, 15°dorsal ele-vated position and 30°dorsal elevated position.The data were collected for correlation and regression analysis. Results The CVP value increased with the elevation of body postures,the CVP value under supine position was linearly positively correlated with the other two positions(P<0.01).The regression e-quation of supine position and 15°dorsal elevated position was:Y=1.009X1+0.811; the regression equation of supine position and 30°dorsal elevated position was:Y=1.005X2+1.630. Conclusions Through moni-toring the CVP value under 15°dorsal elevated position and 30°dorsal elevated position,and substitute it in equation,we can evaluate the CVP value under supine position.

ABSTRACT

Objective To evaluate the long-term molluscicidal effect of suspension concentrate of niclosamide (SCN) spraying in marshland regions of the Yangtze River. Methods A marshland with Oncomelania snails in Yangzhong City, Jiangsu Province, was selected as the study spot, and a dose of 6-8 g/m2 of SCN was sprayed yearly for four successive years, and the status of snails was investigated. Results From 2004 to 2007, the average densities of living snails were 6.00, 4.25, 2.04, 1.95 snails/0.1 m2, respectively before the use of molluscicide, and 0.86, 0.86, 0.23, 0.16 snails/0.1 m2, respectively after that. The average densities of living snails before and after the use of molluscicide reduced year by year, and the reduction rates of average densities of living snails were 85.67%, 79.76%, 88.49% and 91.78%, respectively from 2004 to 2007. The rate of frames with snails after the use of molluscicide was 73.46% in 2007, with a reduction rate of 86.85% compared with 9.66% of that before the use of molluscicide in 2004, and the average density of living snails reduced from 6.00 snails/0.1 m2 to 0.16 snails/0.1 m2, with a reduction rate of 97.33%. Conclusions After the spraying for four successive years, the density of snails reduces significantly and the molluscicidal effect is stable and reliable. SCN is a formulation of molluscicide for long-term application in marshland regions.
